Freedom™ ORF clones are collections of high quality sequence verified
genes cloned into vectors that are free from the IP constraints
commonly found with other recombinational cloning systems. Each clone
has been amplified and sequenced to ensure the highest quality standard.
BD Creator™ Universal Clones
BD Biosciences Clontech Signs BD Creator™ Licensing Agreement with Open Biosystems.
Open Biosystems offers a set of over 1,600 human ORF clones, the BD
Creator™ Universal Clones, which represents the initial release of the
Freedom™ ORF collection. The BD Creator™ system is a flexible
recombinational cloning system based on Cre-loxP reactions. These ORF
clones are provided to you in the pDNR-Dual vector, allowing you to
move the inserts directly into any expression vector of choice through
a simple Cre recombinase reaction. Although Freedom™ ORF clones can be
subcloned into any expression vector using traditional restriction
enzyme and ligase cloning methods, the Creator™ System allows the
target gene to be rapidly transferred in-frame to a variety of
expression vectors. The BD Creator™ Universal Clones are constructed
for immediate 3' tagging with a 6xHN affinity tag.
A large
portion of the genes in this collection are represented by two clones;
one with a stop codon and one without, giving you the freedom to
produce native or fusion proteins to tag the ORF.
Freedom™ ORF
clones are available in two different formats: glycerol stocks and
IsoCode® cards. Freedom™ ORFs spotted onto IsoCode® cards give you the
freedom to store the clones at ambient temperature and use them
directly in PCR, transformation and sequencing. These features, in
combination with the high quality of the clones in the Freedom™ ORF
collection, bring you advantages far above the competition.
The
BD Creator™ System transfers a gene from a specialized Donor vector
into any Creator™-compatible expression vector, called an Acceptor
Vector. The Creator™ System reduces the time required to construct any
number of expression clones to a 15-minute reaction mediated by only
one enzyme-Cre recombinase. The Cre-LoxP recombination reaction
transfers the Freedom™ ORF directionally to the Acceptor vector without
the loss or addition of sequence. The resulting expression clone
contains the ORF in both proper orientation and reading frame. A
variety of BD Creator™-compatible expression vectors are commercially
available or you can adapt the vector of your choice to the BD Creator™
System using a 161-bp cassette. No licensing fees are required to use
this system.
Shipping Information:
Orders for individual Freedom™ ORF clones are provided as a
ready-to-use live culture in a 2ml tube. Each tube contains LB medium
supplemented with 8% glycerol and the appropriate antibiotic. Within 2
days of receiving your order we will ship your clone at room
temperature via express delivery. Store the stock clone at -80°C.
Orders of 50 clones or greater or orders for the complete collection of
BD Creator™ Universal Clones are provided in 96-well microtiter plates.
These will ship on dry-ice via overnight delivery and should be stored
at -80°C immediately upon receipt. Please contact our customer service
department for a quote and estimated shipping time on bulk orders.
Clones on IsoCode® Cards are sent as 3 punches in a 2ml tube. Following
a simple protocol, the DNA from punch is eluted in 20µl of sterile
distilled water and is sufficient for several reactions. These punches
are shipped at ambient temperature and can be stored at ambient
temperature short term. Details for long term storage are included with
shipment of the product.
