Human LentiORF Clones
For transient or stable expression of tagged human ORFs in mammalian cells

The Center for Cancer Systems Biology of the Dana-Farber Cancer Institute has created a large and growing collection of human open reading frames (full-length cDNAs with UTRs removed) derived from the fully sequenced Mammalian Gene Collection (MGC)1. Previously available as recombinational entry clones, these ORFs are now available in lentiviral expression vectors that incorporate specific fusion tags. pLEX vector
Each construct contains an ORF with fusion tag(s) under control of a CMV promoter for expression in mammalian cells. The vector backbone is designed for transient or stable transfection and for lentiviral production and subsequent transduction in a target cell line.

Please note that the pLEX vector is not compatible with third generation packaging systems such as ViraPower™ from Invitrogen. We recommend the Trans-Lentiviral™ Packaging System for use with our vectors.



Two alternative fusion tags
pLEX-HA-MYC codes for the hemagglutinin (HA) epitope at the ORF protein N-terminus and the c-Myc epitope at the ORF protein C-terminus. These small tags are ideal for cell imaging, immunoprecipitation, and studies of protein processing and trafficking.

pLEX-JRED codes for the red fluorescent protein JRed at the ORF protein N-terminus. JRed is not cytotoxic, does not form visible aggregates, and is a well-validated fusion partner for cell imaging. It is also useful as an expression indicator in gain-of-function screens with a GFP reporter.

Individual clones:
Clones are provided as a live culture in a 2-ml tube. Each tube contains LB medium supplemented with 8% glycerol and the appropriate antibiotic. Within 2–3 business days of receiving your order we will ship your clone at room temperature via express delivery. Store the stock clone at –80°C.

Clone Collections:
Orders of 50 clones or greater and stock plates will be provided in 96-well microtiter plates. These will ship on dry-ice via overnight delivery and should be stored at –80°C immediately upon receipt. Please contact our customer service department for a quote and estimated shipping time on bulk orders.
