Validated GIPZ shRNAmir cancer gene set
Bench validated in cancer cell lines
The validated GIPZ lentiviral shRNAmir cancer set was developed with funding from the National Cancer Institute (NCI)  to test and validate shRNAs targeting a set of high priority human cancer genes. After validation, the sequences of the cancer gene-associated shRNAs have been deposited into the NCI's Cancer Genome Anatomy Project (CGAP) open access public database and are commercially available through Open Biosystems. The validated shRNAs will serve as tools for researchers to target and modulate the expression of well-known cancer-associated genes.

Highlights of the set include:
  • Targets important cancer specific genes with three shRNA per gene
  • shRNAmir design increases specificity and knockdown efficiency
  • Bench validated in two cancer cell lines
  • Efficient  knockdown at low copy
  • Guaranteed* knockdown

All the GIPZ shRNAmir included in the validated cancer set were validated at low multiplicity of infection (MOI= 0.4-2). The level of knockdown for a shRNAmir to be considered successfully validated was set to ≥50% of mRNA expression. Over 80% of genes tested had at least one shRNAmir that produced
50% knockdown.

The validated GIPZ lentiviral shRNAmir human cancer set is available as individual GIPZ shRNAmir, target gene sets, or the entire validated set. Only those shRNAmir that produced
50% knockdown in at least one cell line have been included in the set.

Shipping Information:
The validated human GIPZ lentiviral shRNAmir cancer set is provided in 96-well microtiter plates containing frozen stock cultures of E.coli in LB (low salt) broth with 8% glycerol, carbenicillin (100ug/ml) and zeocin (25ug/ml). Individual shRNAmir constructs are shipped as glycerol stock cultures on wet ice. Open Biosystems checks all cultures for growth prior to shipment. Store at -80ºC.
