TRC lentiviral shRNA library

The RNAi consortium (TRC) is a collaborative effort based at the Broad Institute of MIT and Harvard, and includes six MIT and Harvard associated research institutions and five international life sciences organizations. Over three years TRC will create lentiviral shRNA libraries targeting 15,000 human (TRC-Hs 1.0) and 15,000 mouse (TRC-Mm1.0) annotated genes with a goal of validating each shRNA in multiple cell lines. Open Biosystems has partnered with the TRC to make these shRNA libraries available to researchers worldwide.

Features of the TRC shRNA libraries include:

  • Rules-based shRNA design for efficient gene knockdown
  • Already cloned into lentiviral vectors
  • Amenable to in vitro and in vivo applications such as the creation of stable knockdowns
  • Lentiviral vector enables transduction of primary and non-dividing cell lines
  • Broad coverage: 4-5 constructs per gene

Finding your shRNA of interest

  1. Enter gene symbol(s), GenBank accession number(s), clone IDS, Unigene clusters or catalog numbers into the gene search or  the shRNA search in the purchase grid.
  2. Results will be returned in a tabbed format. Click on shRNAs under the shRNAs/RNAi tab.
  3. Click on the oligo ID link for additional information including hairpin sequences.
  4. Order directly online. Constructs are shipped in 48 hours.

Citing the library in Scientific Publications

In scientific publications, the libraries should be referred to as TRC-Hs1.0 (Human) and TRC-Mm1.0 (Mouse). Individual clones are uniquely identified by their TRC ID number (eg. TRCN0000014783).

Shipping Information:

The TRC lentiviral shRNA libraries are provided in 96-well microtiter plates containing frozen stock cultures of E. coli in 2XLB broth with 8% glycerol and carbenicillin (100ug/ml). Open Biosystems checks all cultures for growth prior to shipment. Individual shRNA constructs are shipped as glycerol stock cultures on wet ice. Store at -80ºC.
