Cell Culture

Hybridoma cells are thawed and grown under standard protocol conditions.

Monoclonal Production

Hybridomas are grown under standard protocols until harvest.  Each liter yields 5-20mg of antibody on average, and projects take roughly 4 weeks for completion.   As each hybridoma is different, the final success including the amount of antibody produced cannot be guaranteed. After production, the monoclonal antibody is purified by protein A/G resin.

Ascites Production

Primary immunizations take place with a standard number of cells (or cell number dictated by customer) with the requested number of BALB/c mice (default strain). Approximately 2 weeks after cell inoculation, ascites is produced and collected.

Monoclonal Purification

Once the desired amount of supernatant has been produced, one of several types of purification may be selected. The preferred method for most customers is to use a protein A or protein G resin, although anti-immunoglobin purification can be done as well.  Antibodies can also be purified under standard non-GMP or full GMP conditions as required.  Monoclonal supernatants are purified under high salt conditions for stringent binding
pending the antibody species and isotype. When the affinity of the antibody for an indicated capture resin is not known, small-scale pilots can be performed at additional costs.
